No it can't - unless you count things like the separate trees or (separate) quick view as panels.

Unfortunately almost all functions in Total Commander rely on exactly 2 panels, one source and one target directory. Without this, it would be much harder to use the program with the keyboard, similar to Explorer.

To compensate this, you can quickly change directories with Ctrl+D (directory hotlist) and Alt+Cursor down (history). Total Commander also support directory tabs (as in Chrome or Firefox). You can open them with Ctrl+T, and switch between them with Ctrl+Tab and Ctrl+Shift+Tab.
